You should be able to get tickets from Assisi or Siena. Try the SENA bus company: www.sena.it for information about Assisi-Siena buses. I've never done this route though, maybe someone else here has and can offer better advice? I'm not sure about this, but you might have to get a bus to Perugia or to Santa Maria degli Angeli (where the train station for Assisi is) to change to another bus for Siena. For Siena-Florence, just hop on a SITA bus from Piazza Gramsci, there are many running each day, the ticket offices are in the underpass in the Piazza. The SITA website has a timetable for this route.

Marietta, Rick Steves does an excellent job in his Italy book covering all the details about how and where to get the bus tickets for these two trips, and where to catch the bus.

You will not need tickets prior to your trip. When we did the Siena to Florence trip we just bought our tickets the day of the trip.

Thanks for the info! I have the RS Italy book....and most of the conversation, seems to be about local buses moving about Assisi etc. But I guess these are the same heading from one city to the next?

There was a similar Assisi-Siena bus question about a month ago. My recollection is that there was a morning bus departing from near the train station and a late afternoon bus departing from the big parking lot near Bascilica of St. Francis. The TI office would likely have current schedules and may sell tickets.
