My husband, daughter and I will be there only one night this May. Any of the cities better for finding a hotel or B&B than others? Any suggestions for moderately priced, comfortable place?

If you're only going to be there one night, I'd also suggest Monterosso al Mare. It's the largest of the five towns, and has the best selection of lodgings and also the nicest beach.
Check Rick's books for suggestions on possible places to stay. If you choose something in the "old" part of town (through the tunnel), the cost might be a bit cheaper?

Roberta: We stayed at the B&B Marisa in Monterosso. Very nice and clean and a short walk to the train. The website is www.bed-and-breakfast-marisa.it.

I was surprised as to how many places were already booked for May. After calling dozens of places that rent rooms in Vernazza (we wanted rustic), I finally reserved the room at Caterina's - http://www.caterinarooms.it/. Looks interesting! I'll let you know.
