The Basilica of Santa Maria has wonderful mosaics.
Chef Andrea teaches a cooking class that is loads of fun, even for a novice.
And the walk across the bridge in the evening to get to Trastevere is always lovely.

This walk and the Jewish Ghetto walk start in the same place so we did the Jewish Ghetto walk which is short and then the Trastevere walk followed by pizza at the end of the Trastevere walk. This was one our favorite days we've had in Rome. We've since learned to bring a splitter so we can share 1 iPhone and not have to worry about staying in sync on the audio.

We did an eatwith.com dinner with Federica and Barbara that was a highlight of our most recent trip to Italy. Smack in the center of Trastevere. We had a blast and I now recommend it to anyone going to Rome. More info here:

I second Dar Poeta...but save room for their Hazlenut Stromboli. Hang out in the main Piazza (Santa Maria) in the evening.
St. Cecilia--wonderful ancient church and more recent church built above it.
We did the same eatwith dinner that Michael mentioned above.
It was the most expensive dinner of our trip, but it was also one of the most enjoyable.
We had a wonderful dinner at Dar sor Olimpio al Drago and another at Ristorante all Arco di S. Calisto.
Trastevere has become quite popular, so I'd recommend reservations for any place that you really want to go for dinner.
