Planning a mini vac to see Venice, Florence Milan then a conference in Mannheim have about 5-6 days any suggestion?

Wow, that's a lot to cover in 5-6 days, as well as getting to Mannheim. As Nigel mentioned, a few more details would be helpful. For example, where are you flying from and have you travelled in Europe before?
My initial impression would be to fly into Venice, train to Florence and then head up through the Brenner Pass to Munich and then to Mannheim (skip Milan). I'd have to spend some time working out the details.

I too suggest dropping Milan. 2 nights would be a minimum in each of Venice and Florence. That leaves one (or two) more nights - perhaps choose Padua and/or Verona for stops in between.
Trains are quick and frequent between all these locations.
