Your visit to the Last Supper is time-limited, so you can also visit something later in the afternoon.
Repends on how much time you want to spend at the Castello, beautiful grounds and good museum. I would allow at least an hour for the Duimo, two if you take the elevator to the roof. You will be out of the Cenacolo (Last Supper room) by 2:30, so you could visit one site before the Last Supper and one after if you tend to dawdle (I do).
Diana, you also ought to take at least a few minutes to wander through the Galleria right near the Duomo, even if you don't take any time to go into any shops or grab a beverage or snack. It's really impressive and lovely, and certainly free to wander, although no doubt expensive if you actually shop!
