I had originally planned to be in Naples for the day on May 1st. Since that is a holiday would it be better to schedule another day there since some places might be closed? Do things close up?

What places are you interested in? If it's indoor sights like museums, the easiest thing to do is check their websites. Normally, holiday closings are shown there along with the basic days and hours of operation.

A lot of restaurants are closed in Napoli on Sunday, even without a holiday, but I plan to be there on May 1 and am not concerned. I do know that Sundays are quiet generally in comparison to other days.

It's Labor Day, Italian style. You could just decide to enjoy it--you'd get to see people relaxing, walking around with a gelato with their kids in tow. People window shop, have picnics, etc. I was in Milan once on 1 May and thoroughly enjoyed myself.
