We have a half day free where we could spend it in one of these three areas. I'm looking to stroll around, maybe eat lunch, stop in the shops, and just take in the atmosphere/culture..We are staying at Masseria Salamina, Pezze di Greco and have a car. Which would you recommend?

I have not been to Martina Francia so I can't comment on it. However, I rented a trullli at La fogliarella resort in Cisternino. I would recommend Ostuni because it is agreat town with shopping, restaurants and sights. Locorutundo is smaller and there is less to see and do.

We loved Martina Franca and did not have nearly enough time there. Beautiful town, but be aware that there is a definite pausa from 13:00-16:00, so arrive in the morning, have lunch at Italian time (about 13:00) and then move on as nothing will be open.

As I recall, all three shut down in the afternoon. My favorite is Ostuni.
