Flying out of Marco Polo at 6:30am. I’ve read co flirting things that the airport isn’t actually open before 5am? I’m staying near the airport at Best Western Titian Inn Hotel Venice airport. I see they offer a shuttle to the airport starting at 5am. Is that going to get me to the airport in time? When should I arrive to the airport? Other options for getting there?

Your flight must be going to another European city so 2 hours in advance is enough-think of it as more like a domestic flight
You will exit Schengen at whatever city your flight to US departs from
You can call your airline and ask them what time check in desk will be open
If you must be there by 4:30am have your hotel arrange a taxi

Yes, I think that will do. It’s been a few years, but I’ve stayed at that Best Western and they’re good. As I recall if you’re there at dinner time there is a surprisingly good restaurant within walking distance.

You will exit Schengen at whatever city your flight to US departs from
UK and Ireland are part of "Europe" but are NOT Schengen countries.
If your flight is going there, then you will need to do passport processing in Venice to exit the Schengen zone.
Christine's comments are correct, IF that departing flight lands at a Schengen zone destination.
