I am planning a trip for 16 of us in September. We first of all need to pick pick which lake and town. Please realize that when we leave here we are headed toward Barolo and don't want to drive forever. I am leaning toward Lake Como, Where should we stay? (hotel and town?) Any suggestions in either area are greatly appreciated.

Then you should go to Lake Maggiore. Closer to both Barolo and Milan Malpensa airport (MXP), where you are likely to land if you fly to Milan.
Stresa is a popular town on Lake Maggiore and very convenient for the ferries to the islands. I presume you will be renting cars. Barolo is a small village in a rural part of Piedmont called Langhe, a famous wine producing area, and therefore not served extensively by public transit.
