May 20th-traveling from Rome Airport to Cortona by train May 27 Cortona to Cinque Terre by train May 31 Cinque Terre to Rome Airport by train. What is the quickest route by train to these places?

If you enter your departure and arrival cities on Trenitalia.com, it will show you the routes. Pick a date six days from today to see prices, but your trips involve regionale trains which can't be purchased this far in advance.

To add to what Zoe said when you check for tickets on Trenitalia set the time for the ticket for early in the day, say 7 AM. If you leave it alone it will give you trains at the current time and after which will then appear to offer limited choices.

There are two airports in Rome. If your flights are to/from the US, most likely you mean Fiumicino.

Regarding timing:
Fiumicino to Termini, main train station in Rome, is 30 minutes-ish

Termini to Cortona is 2 1/2 hours ish

Cortona to La Spezia (the main city that is considered the "entrance" to the towns of the Cinque Terre) is 2 1/2 hours-ish

The trains between the CT towns are very short rides, 5-15 minutes depending on your stopping/starting point

La Spezia to Termini is 3 hrs 45 minutes if you get one of the few direct fast trains every day. But if you get on a train that makes frequent stops, this could be longer than 5 hours. For this reason, I would recommend leaving La Spezia the day before you need to catch your flight from Rome, unless you take the first train out in the morning and have a later evening flight out of Rome.

As the others suggested, you can look on the trenitalia website and enter your dates so you can see the train times and which trains you would want to take. You can buy the tickets starting from 120 days out and this saves a lot of money for certain trips although does lock you into a specific time and train.
