We are visiting Cinque Terre and Florence in early October. We will be driving from Florence to Rome for our return flight and want to make stops along the way. We will have 4 nights to spend along the way with the last night staying near the airport. We have previously been to Siena, Rome, Assisi, Venice, Pisa, Amalfi and Capri.
Any recommendations would be appreciated.

I’ll second the suggestions for the Montepulciano area. If you are into wine at all you’re spoiled for choice among some great wineries in the area. Awesome scenery, good food, and some relatively less crowded spaces.
Pienza, Montalcino, Monticchiello are worthwhile visits and easily drivable from Montepulciano, as is the A1 for your return to Rome.
I liked Orvieto a lot. Rick covers it fairly extensively in the guide to Italy, though unfortunately not in the book on Rome.
