Monterosso is the most convenient town when going by train since you can connect once. The other towns require multiple connections.
The fastest way is probably a rented helikopter. The best way will depend on your personal preferences.

Venice > Florence > Pisa > La Spezia > any of the CT towns, will probably be the route you end up on.
We traveled that route by train; Venezia Santa Lucia to Monterosso; approximately 5 1/2 hours. Easy transfers.
How many train transfers? According to Treniitalia website I would have to change trains twice and it takes 7 hours.

Ditto to the above. We need to know if you're leaving from Venezia Santa Lucia station, which of the 5 villages in the CT you're trying to get to, and the date of travel. All of this makes a difference.
In any case, the CT is a bit of a haul from Venice.
leaving Venezia Santa Lucia on October 4 to Vernazza.
Also leaving Vernazza on Oct. 7 to Naples.

There are several solutions
All will take anywhere from 6-7 hours
All require 2-3 changes
You are crossing Italy at its widest point and heading thru an area not well served by fast trains
You can see where each change is on the Trenitalia site by opening the Details (Changes) and Stops for each leg
The Vernazza to Napoli trip is basically the same-6-7 hours, 2 or 3 changes
I you choose via Genova (Genoa), make sure you aren't changing stations there.
Via Milan you can go from Venice to Monterosso in 5 hours and 30 min. One change and reserved seats all the way to Monterosso, but getting to Vernazza from Venice will always involve one more change.
There is no local demand for direct, subsidised, IC services from Trenitalia whereas the lack high speed tracks between Milan and Genoa makes such a route unprofitable for Freccias and Italotreno.
