I read in Rick's book that you can bypass the line at the Vatican if you prebook a guided tour by faxing the vatican. I did so, but the number in the book would not work. The Vatican web site has another fax number, but no reply.

If you search around here, you will find many threads where people say it took weeks for the Vatican to reply. Some people here and on TripAdvisor have said that they didn't know if they got a tour until they arrived at their hotel in Rome, which is where the Vatican sent their reply. So it may take a while before you hear back. Good luck!
