What is the best ( economical) way to travel from Venice train station to Hotel Rio

Edit: Whoops! I misread and answered the wrong question! Definitely Vaporetto, as Lola says.
It looks like an easy walk from the San Zaccaria vaporetto stop. Line 1 from the train station would be easiest, heading right down the Grand Canal.
The Alilaguna is for transport from the airport, not the train station.

From “Piazzale Roma” Bus Terminal
Catch ACTV water-bus n. 5.1, which leaves from dock “E”, and get off at “San Zaccaria” dock “F”, in front of Danieli Hotel. From that point take Calle delle Rasse (a small alley next to Danieli Hotel) or Calle degli Albanesi (the parallel one) and continue straight until you reach "Campo SS Filippo e Giacomo", where our hotel is located. This line runs through the "Giudecca Canal" and the whole trip takes 20 minutes.
Alternatively it is possible to catch ACTV line n. 1 from "Piazzale Roma" dock “D” and get off always at "San Zaccaria" dock “F”. This line runs through the "Gran Canal" and the whole trip takes approximately 43 minutes.
At night catch ACTV line “N” from dock “F” and get off at "San Zaccaria" dock “F”.
Unfortunately those directions apply to transport by vaporetto from Piazzale Roma, not the train station. The hotel is assuming that people are arriving at the airport and take the bus to Piazzale Roma.
Those directions will not work from the train station (Ferrovia on the vaporetto schedules and map). They would have to walk down to the Ponte della Costituzione and cross over that that bridge; not something I recommend with luggage.

From the train station, they need Vaporetto Line 1, which goes down the Grand Canal to the San Zaccaria stop. (Line 2 also goes down the Grand canal, but not as far as San Zaccaria docks, so they would have to walk along that section which is the most crowded sidewalk in Venice).

To take any vaporetto, one needs to know which dock serves that particular vaporetto in the correct direction. According to Moovit, Line 1 toward San Marco/San Zaccaria uses Dock E, which is toward the right as you face the Grand Canal from the train station.
You must buy a ticket AND validate it at the machine before boarding.
The Line One instructions in Mary's post work fine, just understand that Ferrovia (train station) is one stop after P.le Roma across the canal. Simply come out of the train station and board the Number 1, destination Lido, and follow the above.
To make it most economical, instead of paying the very high single ride prices, buy a pass for the duration of your stay. They pay for themselves very quickly. Remember to scan in every time before you get on the boat.
