If we use the HOHO as a way to orient ourselves to the city, will it keep the attention of 7 & 10 year-old boys or will it be too much?

Since the point of a hoho bus is you are not stuck on it , but can get off and on as much as you like, at any site , it is not too much for children at all. Just get off, look around, get back on and go to next site.
I think the on/off buses are perfect for kids. Get off, walk around, find a bathroom, get back on. Far better than being trapped in a bus for two hours. Many of the tickets are good for 24 hours so buy about noon and use it for two days. One other thing we often do is the completely ride the loop one time just to get an orientation and a feel for what we might like to see longer. On the second loop we get on and off.
I agree with the above posts. The #110 city bus tour covers over 80 sightseeing destinations. The ticket is good for 24 hours w an audio guide. Sit on top in front for the best view.
Follow Jeff's advice. And stay busy pointing out sights along the way which will interest your boys.
Get off as often as you need to.
Lisa - You may also want to check out the Archeology HOHO bus, which can be picked up at Termini (or elsewhere). Takes you to the aqueducts, Appian Way, catacombs, etc. My wife and I enjoyed it and I think it offers some attractions the boys will find interesting. Good luck :)
