Does anyone know a local tour guide in Florence who would take someone on a day trip to Pitigliano? I am really interested in this town and am researching all possible ways to get there other than driving myself.

I can't answer your question directly, but I had a thought for a research direction. Pitigliano, before WWII, had a relatively large Jewish population. In fact, it was known as "Little Jerusalem." It is now on the Jewish tourism route. Maybe you could find a guide by googling Italy, Jewish tourism or by posting a question on the Tripadvisor forum.
BTW, have you read Edda Servi Machlin's, "The Classic Cuisine of the Italian Jews?" Lots of information about the town and Jewish life there, as well as recipes.
