What do you recommend as the best way to get from Siena to Manarola w/o a car?

Train from Siena to Empoli (on the Siena-Florence rail line).
At Empoli change train. Train from Empoli to Pisa.
At Pisa change train. Train from Pisa to La Spezia.
At La Spezia change train. Train from La Spezia to Manarola.

The route described by Roberto is about the quickest way to travel that route by train, with a travel time of about 3.5 hours. There are some trains with one fewer change, but slightly longer travel time. Check the Trenitalia website for a description of all the trains on that route, departure times, prices, etc.
Many of the trains will be Regionale, so DON'T forget to validate your tickets prior to boarding the train on the day of departure. I'd suggest buying your tickets prior to the day of travel, perhaps when you arrive in Siena.
