Does anyone know the best way to get from the Amalfi Coast (will probably be staying in Positano)to the Tuscany region? We will not be renting a car. Thanks in advance for any help you can provide!

Where in Tuscany? Without a car, you're limited to towns along the major train routes. Florence is perfectly reasonable.
But to get you close, I'll tell you how to get to Rome. From Positano you'll take the SITA bus to Sorrento. Uphill (about 40 yards) from the bus stop is the station for the Circumvesuviana train to Naples. About an hour to Naples where you'll go upstairs to catch the train to Rome - about 2 hours.
From Rome, I'd need to know where in Tuscany you'd want to go. Florence is about two hours.
