Of course you can, there is no law against your plan.
You can rent a car at the following Tuscan locations which are easily reachable by train from Rome: (in order of appearance)
CHIUSI-CHIANCIANO
AREZZO
FLORENCE

You can rent also in other cities, but the above are the cities with direct (no changes) train connections from Rome.

Hours Google for Wandering Italy Val d’Orcia, southern Tuscany. Drive 2.5 hours from the Roma Termini station, or trains to Chiusi at 7:38 and 9; Hertz half block from the station [closed sunday].

What we have done several times is take the train from Rome to Chiusi. Just outside the Chiusi station is a Hertz to rent a car. Keep in mind that they close at noon on Saturday and are completely closed on Sunday.

Once you have a car, I suggest heading to Montepulciano as a base to explore from. It's about 30 minutes from Chiusi. Montepulciano is a beautiful town and probably our favorite town in this area of Tuscany and in the epicenter of wine country. Amazing restaurants, shopping, festivals, views, wine, etc...

For accommodations, check either Charming Homes (charminghomesmontepulciano.com) for an apartment in the historic center or Fonte Martino (fontemartino.com) for a B&B experience just outside of town.

Hope this helps!
