For Galleria degli Uffizi and Galleria dell' Accademia. Is it necessary to reserve advance tickets to these two museums in September? Are they still crowded during early-mid Sept?

The short answer is YES, they could still be very busy in September. My last visit to Florence was mid-September, and I observed long queues in the "tickets" line at both sites.
The owner of the Hotel I was staying in kindly made reservations for me so I didn't have any problems. You could "take your chances" but I'd suggest reservations would be a good idea.

Same advice as Ken, which is the standard or usual advice given here on this question.
September is not really shoulder season in Florence, somewhat shorter lines than the summer.
Reservations eliminate the downside possibility of having a long wait in line, makes your schedule for that day(s) more predictable.

As easy as it is to make reservations I would do so. I just returned 2 days ago from Italy and we were so glad we had reservations for the Accademia and Uffizi. If I had not made them we would not have gotten in. I called to Florence several weeks before we left home and made our reservations. You can go to their website and see the available times and then give them an exact time when you call. I know people were having trouble booking them online through the official museum website so I decided to call instead.
Jenny we went last Oct. and the lines were long,get in touch with your hotel and tell them what day and the approximate hour and they will gladly make the res. for you ,the same in Rome,Sienna etc.If you want to see the Last Supper in Milan reserve a tix. online maybe a week or two ahead of your visit, have fun,you will get your fill of Madonnas and Jesus but still great,the David at the Accadamia is the best
Definitely get your tickets in advance to both museums. It is worth it. You don't want to spend your precious time waiting in line. Both museums are worth the visit. Also, if you have time, visit the Bargello and don't miss the Duomo Museum. RS recommends both.
