You could go that. Assisi is such a beautiful small city.
The trip to Positano will take time, and personally, I wouldn’t do it. But I wasn’t enamored with the town.;I like Ravello the best on the AC.

The best day trip from Florence is Siena by direct bus (no transfers) that will drop you off outside the old city, Don’t take the train because it takes longer and then you have to take a bus from the train station to Siena’s old town. The bus takes 1h 15m.
Good day trips from Florence’s SM Novella station by direct train: https://www.trenitalia.com/en.html are:
Venice but get an early start and get off at the Santa Lucia station that’s the end of line meaning get off when everyone gets off (2h 15m).
Rome and get off at the Rome’s Termini station (1h 45m).
Milan’s central station (2h).
Orvieto and then take a funicular to the top of the town (another place you want to get an early start [2h 15m]).
Padua (Podova) (1h 45m) reserve tickets for the Scrovegni Chapel before going.
Pisa (1h)
Lucca (1h 30m)
Will you be able to rent a car some days? If so, Florence is a great jumping off point to visit the Chianti wine region along the SR222 'Chiantigiana Road': https://lovefromtuscany.com/scenic-drives-in-tuscany/.
Other good day trip options by car from Florence are:
Volterra (1h 30m).
San Gimignano (1h 30m).

zimmermanmc, I'll refer to a similar question you asked about a week ago.

https://community.ricksteves.com/travel-forum/italy/italy-for-one-month

You said:

Now I am thinking about 2 weeks in Florence the perhaps 2 weeks in
Spello. I have been to Florence, Rome, Pienza, Montepulciano and
Cortona and Sienna.
Also visited the Amalfi Coast. This time i would
really like to experience the local life in Italy and share it with my
daughter. Prefer not to stay in a hotel. Any suggestions would be
great!!!

Suggestions were contributed that post. As you are already familiar with Florence and the Amalfi Coast, I'm a little unsure what sort of help you are looking for? Can you do Florence, Assisi and Positano (which doesn't have train service, BTW, and no ferry service in the early spring) in a month? Sure. Can you find enough to do in and around Florence to keep you occupied for 3 weeks? Would you like Positano enough to make the long trip from Florence or Assisi worth the time? Depends on your interests. What can you tell us about YOU (and your traveling companion, if bringing one along) that might help us best answer your question? :O)
