Five nights Cinque Terre

For a future trip, I'm thinking of five nights in Cinque Terre, one night in each village, hiking from one village to the next. Is this practical? Or too slow-paced? Is the second half of April a good time to visit?

It is just not necessary to change accommodations, as the villages are minutes apart by train. You can also use ferries (seasonal), so pick one base and hike as much as you like, supplement with public transport when you don't.
I'm an avid hiker, but three full days would do it for me. Nothing wrong with a slower paced trip though, if that is what you want.

Three nights eh? We are a middle aged couple and would enjoy a quiet village at night. Do you have a recommendation on which village to stay in?

I do not, but there are many recent discussions of pros and cons of various villages and specific recommendations--I would enter Cinque Terre and accommodations into the search bar.

We stayed last October in Manarola. We stayed at Ca' Andrade (I may be spelling that wrong). It is in the RS Italy book.
Manarola is very quiet at night. From there we did have to train or bus to the starts of the hikes as the hike between Manarola and Corniglia was closed, but this was easily done. One of our favourite hikes was taking the bus up the hill to a town called Velastra and hiking down to Corniglia where we had a drink and snack. Spectacular views and very few people doing that hike.
