With just one day (or parts of 2 days?) in Florence, I’m guessing you’re planning on seeing sights just within Florence on Days 4 and/or 5. But for Venice and Rome, are you looking for suggestions for trips outside those cities, or strictly inside the cities?

Buy Rick Steves guidebooks for those areas and you cant go wrong, Download his audioguides - free! and again, you cant go wrong. There is SOOO much to see you will want to go back, I will recommend RSs Ostia Antica tour.
Book as much as you can in advance to save you precious time and aggravation
Enjoy!!

What are the interests of the folks in your group? Is this an organized group that will go everywhere together, or is family members, or a group of friends, who may split up to do different things sometimes.

There are lots of choices in each place. There are "top sites" which are the most popular, but there's no reason to go if something else sounds more interesting to you. What's the dynamic in your group - is everyone ready to let you determine the itinerary or do you have to get consensus?

Moving that many people as a group is challenging. Florence is all walking and in Venice there's almost always going to be room for everyone on the vaporetto, but in Rome, you may not all be able to get on the same bus. You'll probably need 3 taxis if that's your choice in Rome. Even walking as a group can be very slow . . . in a guided tour, everyone wants to keep up with the guide and always knows where s/he is. An informal group is much less motivated. Just sayin'
