As I recall, when you walk down the front steps of Venezia S. Lucia station, there's a ticket booth just to the left. You'll be able to buy Vaporetto tickets there. One point to mention is that you must validate tickets before boarding the Vaporetti. The validation is electronic and a bit different than the trains. Touch the card (ticket) to the reader and if you get a green light, you're good to go. DON'T forget or you may face hefty fines!

At the station you can buy the passes both from the indoor ticket booth to the left - which also sells tours, toilet tickets, maps, etc., - but also from the little ticket booths along the water right at each dock. There are also small ticket booths at most major stops, but many minor ones only have machines.
My experience over many years is that the people over the face to face counter are much much more helpful than the ones in the boxes behind the thick plastic window. They all speak English, too - unlike the ladies in the little boxes.
Remember to tap in every single time, get the beep and green light.
I have usually bought my vaporetto tickets and passes at the tabacchi shop inside that train station.

You can even purchase your vaporetti passes on-line direct before you leave home but you'll have to exchange the voucher at one of the ticket offices.
One word of caution... The vaporetti stops in front of Santa Lucia are an absolute zoo most of the day. You are much better off crossing the bridge and getting on at San Stae or even Rialto market. (This also goes from getting on around St Marc's going in the other direction in the late afternoon and evening.) Whatever you do, make sure you are getting in the queue for the doorway closest to the vaporetto loading ramp - get in one of the other queues and you'll never get on board.
