We will be in Florence, then traveling to Cinque Terre for 2 nights. The last leg of our trip is 4 nights in Venice. By train, what would be the fastest route from CT to Venice?

The routes have changed a bit over the past two years. The fastest routes from the CT now route via La Spezia Centrale, Pisa and Florence. You will most likely change trains in Pisa and Florence and possibly a third time at La Spezia. Journey time is just under 5.0hrs. Most likely, you will be on the slower Regionale trains until you get to Florence. The train between Florence and Venice will most likely be a high-speed. There is also the route from La Spezia to Venice via Parma and Bologna. About the same time and number of train changes. But, Trenitalia won't show you this run as they don't have a "via" option as do some other train websites. You have to ticket the La Spezia/Bologna and Bologna/Venice segments separately.
If you go to the trenitalia site it will give you all the options for travel. Just pick the best one for you. Donna
When I was looking for a fast train someone told me to put in Monterosso as the departure point as some trains only stop there and not in other Villages so try that.
It is true that more trains stop at Monterosso than at any of the other villages. However, nothing better than an IC train stops there. As mentioned above, these trains will take you to La Spezia Centrale or Pisa to change trains for Florence (assuming we're still talking about the fastest run to Venice). There are Eurostars out of Genoa going south but they all whiz through the CT to stop at La Spezia Centrale.
