We find ourselves with a free weekend in Italy in late March and are looking for a not too far out hotel that can accommodate my daughter's family (2A & 2K 5&8) and my husband and me (with a separate room)
The best we've found are 2 - 3 person suites, or 3 2-person rooms (which gets pretty $$). Are there any hotels with 4 person spaces?

Have you tried looking on booking.com? You can specify # of people in 1 room/suite. Looking at the last weekend in March, availability is understandably limited, giving the lateness in booking. But some do exist.
Many years ago, family members took their grandchildren to Venice and stayed in a family apartment at PENSIONE ACCADEMIA. They loved it. I have no other details, but you might check it out--it's a very good hotel in dorsoduro, just near the Accademia Bridge.
Look for appartements with 3 bedrooms on Booking.com and VRBO. That's my best suggestion, but if you are talking March 2026, you are awfully late :-(
Hotel Paganelli has a 'family room' with a double and 2 twins. I have stayed there twice and highly recommend.
How about these apartments?
Monastery/convent accomodations usually have 4 bed rooms. We stayed twice at Istituto San Guiseppe. Great location. good price. check it outbat monasterystays.com
