We have a 7:30am flight from Marco Polo airport in Venice on May 31. Any recommendations on where to stay the night before to ensure we make the international flight?
I would suggest Hotel Olimpia Venice - it is right near the bus station to the airport and also very convenient to a vaporato stop. I've stayed there on 2 occasions and it is perfect as both times we had early flights. The hotel on both occasions packed us a "breakfast to go".
I'd be surprised if a 7:30 flight is an international one. It is almost certainly a short flight to a transfer point like Paris, Amsterdam, London or Frankfurt. I don't think any transatlantic ones are that early.

If a connector 6am should be plenty early. Your flight is by no means the first one out of the airport but Venice has a reputation for early flights.
I'd suggest staying in whatever hotel you have reserved, not a special one for your last night. It isn't hard to get to the airport for 6 am. Depending on where you are staying you have a number of options, also depending on how many "we" is. If "we" is a few or several a water taxi will be affordable and fast, door to door - and scenic. If you are close to an Alilaguna stop that takes about an hour. Or the number 1 vaporetto or N vaportetto at night goes every 10 minutes to P Roma where there is a frequent bus to the airport.
Can you clarify you flight and hotel location?
There is a Marriot Courtyard right outside the airport if you're worried about making your flight on time. They have a shuttle bus that will take you to the terminal.
If you have the opportunity to select a hotel, you might look into something closer to the Piazzale Roma. The first bus (ACTV) departs there for the airport at 4:40am. The second bus (ATVO) departs at 5:00am. The next two buses depart at 5:40am. It's a 25-minute ride to the airport. Plenty of time to check-in for your flight. The suggestion here is to avoid a really early vaporetto ride to get to the Piazzale Roma. If you are down near St. Mark's plaza and you want to take the 4:40 or 5:00 bus, you need to take the night vaporetto that stops there at 3:40am. One suggestion for a hotel is the Hotel Papadopoli. It's about 500' from the Piazzale Roma. Nice hotel and you can walk to the bus at 4:30 or 5:30 in the morning.

From Italy to Spain it is within the EU Schengen Zone.
It's an important distinction because those flights are like domestic flights within the USA, therefore the check in requirements are shorter than those for flights to NorthAmerica or other Non EU countries.
