It's actually 5 sites as the duomo itself is free: you'll need to stand in line for that one. Your 72 hours start with the first entry to any of the 5. You are also required to make a pre-reservation to climb the dome. If all slots are filled when you buy your ticket and try to make that reservation, then it's a no-go. For people purchasing today, the site says the soonest available slots are on July 6th.

Ask when you inquire about a tour if no one on the forums knows if it's a different procedure than when you purchase tickets. Whatever the case, I would not wait to try and do this until you get to Florence lest there be no openings left.
