Renting a car at Florence airport and spending a couple nights in Montepulciano. Can I drive to Naples and drop it off there? Or is there somewhere else I’d need to drop it? Then take the ferry to Sorrento as we’re spending a couple days on the Amalfi Coast. Thanks!

The easiest place to stop your rental car off is Naples airport. We drove from Umbria to Naples, all on the autostrada, like our interstates, and easily dropped it off at Naples airport which is above the city. Much less expensive than dropping it off earlier in your travels. Taxi to ferry dock, done!

you will need to see the companies at Florence airport and see which of those have a Naples airport location too. Then check for any drop charges.

Or drop off the car in Chiusi and do the 4.5 hour trip to Naples by train?

I find it much easier to return the car in Sorrento. Hertz, Avis-Budget, Europcar are all in Sorrento. In your case that is what I would do.
