driving through Tuscany from Florence to Rome for food tour questions

Hello All, I have read alot about driving from Florence to Rome through Tuscany on this forum. I heard SR222 is a popular route to take. My goal is to stop by little towns and eat great food in Tuscany region while driving from Florence to Rome. I don't drink wine. My questions are:
Where to pick up my rental car in Florence and drop off my rental car to avoid driving in the city?
Where to stop by for a night in between Florence to Rome as I don't want to be rush with driving?
What towns are must visit on the way to Rome?
All your tips are welcome.

For me the best area is the Val D'Orcia which is basically between Montalcino and Montepulciano on the map.

In general for exploring the scenic areas in Tuscany (definitely best with a rental car)
The road will often go around not through a town, as the town centers are pedestrian only.
The areas that are not town centers are mostly farmland, when you leave the farming area and approach a town, it typically involves finding a place to park and then walking into the town.

This slows you down considerably and brings up things you might otherwise not consider, for example leaving luggage in your unattended car parked far out of your sight.
With the exception of a random winery or agriturismo there will be very little to stop off at other than the towns. Beautiful scenery yes that is abundant and worth stopping for in many locations.

We picked up our car at the Hertz location on Via del Sansovino...it was easy to follow the signs toward SR222. We stayed outside Siena for several days, drove to Montone in Umbria, then returned the car to the Rome airport as we were flying out the next morning. We would do the same if we were planning to stay in Rome...just take transportation into the city from the airport.
