We will be flying into Rome and then driving to Salerno. Any recommendations on routes? Will there be tolls? Should we expect to hit traffic?

ViaMichelin can give you the fastest route, estimated time, and estimated costs. Yes there will be tolls. Yes there will be traffic. Is there a reason why you chose to drive rather than take the train?

The train is probably faster and easier
If you are coming off an overnight flight the train to Salerno is probably a wiser choice

I remember taking a bus from Rome to Naples on a Saturday for an outing when I worked at the Embassy. It was horrible! Traffic intense, took over 3 hours! We later did the trip on a train in a little over 60 minutes. If you've never driven in Italy, be prepared as it is nothing like driving in North America.
Can you take the train and rent your car in Naples, or even better take the train all the way to Salerno? Do you have parking at your lodgings? That can be challenging as well.
