My husband and I will be Domodossola, Italy for a few days. Then taking the Centovalli Railway to Locarno. From Locarno we need to make our way to Tirano. From Tirano we will be taking the Bernina Express to Chur. My question is: can way get from Locarno to Tirano via railway? Many thanks in advance.

Yes.
You can take the Cento Valli back to Domodossola, then take the train to Tirano (one change in Milan).
You can also take the Swiss train from Locarno (via Lugano) toward Milan, but then change at Monza to catch the Milan to Tirano train.

that's a long slow journey, and the Centovalli is no Michael Schumacher either.
From Locarno it is 13 stops and 1h44 on regional trains at 22 past each hour to Monza. 26 minutes in Monza then 9 stops and 2h22 to Tirano.
From Locarno to Tirano is 4h32.
Some day. Where will you stop for an intermediate night?
