Stay 2 nights in Rome. 9/25,9/26,9/27

3 Nights in Positano. 9/27-9/30

(Maybe fly naples to genoa) To get there quicker

2 Nights in Corniglia. 9/30-10/2

(Rent Car now)

4 Nights in Lucca. 10/2-10/6

3 Nights in Pontassieve. 10/6-10/9

Fly out of Florence 12:10 pm on 10/9

Staying in Airbnb or VRBO accomodations and doing daytrips from the aforementioned places. Do you see this being doable? Anything we should see along the way? Great places to eat? Which train/ferry to use. Can we book it ahead of time? Any boats for rent while on the coast?

This works as it looks like you've done your research and based the length of stays on your groups' level of energy and interests.
On the way Rome to Postiano you could take the ferry from Salerno http://www.positano.com/en/ferry-schedule.
You can book ahead of time the train Rome to Salerno. http://www.trenitalia.com/
FYI: It looks like there are 2 flights a day Naples to Genoa one in the morning and one in the evening.
You may know already La Spezia is the place to rent your car on the way to Lucca
