We are in Florence March 24-28 and are wondering about doing a day trip from there. Any recommendations about best places to go ?
I would do Siena (leaving early) as there are so many things to see in Siena.
The other three days will not be too many to visit Florence.
If you do not wish to go too far and want to see how countryside is like, go to Vinci and visit Leonardo's house.
A trip into the Chianti countryside is very nice. I recommend Tours By Roberto (also in the R.S. books) for a combo wine/history/countryside.
Siena is best done by bus, which delivers you close to the city centre. Check for timings at the bus station in Florence, on a side street off the station square.
Alternatives are Lucca and - why not? - Pisa, which can easily be combined on one trip. There are frequent direct trains from Florence to Pisa, and from Pisa (Pisa S. Rossore is ten minutes from the main attractions) to Lucca, and from Lucca back to Florence.
We did a wine tour in Chianti through "Fun in Tuscany", which also visited San Gimignano.
We also did a day trip to Cinque Terre through "CAF Tours".
Both were great, but if you have to do one, I would choose touring the Tuscan countryside.
In March I would probably visit another city - either Siena or Lucca (I loved Lucca) - rather than the countryside or wineries which are much nicer to visit in summer or autumn.
Has anyone taken a Vespa tour out of Florence?

I just got back from Italy last week and while I have been in spring and fall, we thoroughly enjoyed our drives through the Tuscan countryside. End of March should be even warmer. Did a few wine tastings and they were great. Also did a trip to Assisi, and while it was further than I expected we really enjoyed it.... as well as Orvieto...
If you want to get out of a city environment, a trip into the countryside with wine tasting would be a good option (your hotel can help you book if you need a tour). Other cities have been mentioned; I would add Arezzo to the list of possibilities.
The tour company "Fun in Tuscany" offers Vespa tours. I took their Chianti Wine Tour last summer and it was wonderful. I spoke to a few people who did the Vespa tours and they loved it!
