Yes it's possible, and train is the best way to do it. You'll want to make sure you have all of your sights/museums booked in advanced so you can minimize your time spent queuing.
You can look up itineraries on youtube or trave blogs but basically you take an early train from Rome and a late train back.
Yes, this is certainly possible. I've done it more than once. Book a fast train, leave early, plan well, and you'll be set.
I have done the opposite several times; both tourist day trips and business trips leaving in early morning and coming back in the evening. The trick is doing it by train, as a one-way travel can be done in 1 hour 40 minutes (sometimes even a bit less), while by car you are more likely to need more than double the time. The open country landscape in low Umbria and near Rome is quite hypnotic from the fast train windows.
