We will have 2 days in Rome in September. I need recommendations for a day bus tour from Rome to Positano to the Amalfi coast. Thanks.

My recommendation, especially if you only have two days in my favorite city on the planet, is to not go to Positano or the Amalfi Coast at all. Minimum 3+ hours each way by bus, not great sights on the way either.

There are two major issues:
1-There is much more to do in Rome on a first visit than can be done in 2 days
2-It takes so long to get to and back from the AC (tour or self-excursion) that you have little time left for visiting. Google Maps will prove this to you. Using a professional tour bus does not reduce the likely driving time below 5 hours to Positano. Even 4 hours to Sorrento (if you are lucky with road traffic) seems prohibitive to ME.
Don't forget to pre-book whatever in Rome is a must-do for you, like perhaps the Coliseum or the Vatican Museums and the Sistine Chapel. I think even the (modest but important site) Pantheon has started requiring reservations.
