We are currently in Rome and I went on line to get tickets to the Colosseum. However, I do not have access to a printer, so if I take the e-mail to the Colosseum, will I still have to wait in line to get my actual tickets?

Take your email with you to the special events counter to show them. They will print up tickets for you. You won't wait in the regular ticket lines since you already have tickets and just need to pick them up. You'll still need to wait in the security line, which can be long, but there is no way to avoid that regardless of the tickets you purchase.
