We used Viator.com to book a tour of the Colosseum (underground, arena and upper tier). We did not have to wait in line to get in and found our tour guide to be very informative.

You may also purchase the tour online on the coopculture website. You want the "Guided tours in English" option on the page.

http://www.coopculture.it/en/ticket.cfm?office=Colosseo%2C%20Sotterranei%20e%20Terzo%20Ordine&id=0&evento=6

But bookings don't become become available until every third Monday of a month for the following month, so May 15th for June tours. You will also need to purchase general admission tickets in addition to the tour. That can also be done online or at the reception desk when you arrive for your tour. Additionally, if you're planning to visit the Palatine/Forum before the Colosseum, you can buy your general entry tickets at either entry (1 ticket covers entry to all three sites).

Tours sell out very, very quickly: some members have gotten up in the middle of the night to get their reservations in on the first day/hour they became available!

You may also make your reservation by phone: all of the info is on the page linked above. Reservation office hours, Rome time:

Monday to Friday from 9.00 am to 1.00 pm and from 2.00 pm to 5.00 pm
Saturday from 9.00 am to 2.00 pm
+39 06 399 67 700

And, of course, there's always the option of booking with tour companies. "Walks of Italy" has been very popular with RS forum posters, albeit more expensive:

I would definitely recommend the official tours as stated above via the coopculture website. Not much point in paying extra for a third party tour when the official ones are very good.

You do not need to book by phone. They haven't become available yet for June. You'll need to purchase them on May 15th when they become available. Use the official website to book to get the best prices.

Don't forget that you'll also need an entry ticket along with a tour ticket. If you decide to book by phone you won't be able to do that until May 15th also. Just purchase online. There haven't been the widespread reports of people not getting their tickets with this new system and the system hasn't crashed like it has in years past when new tickets became available. Be ready when the date opens up to buy them so you can secure the date you want.

We used Througheternity.com for tours of the colosseum/roman forum as well as another tour of the Vatican/Sistine chapel.

Fabulous tour company. Lucca led our tour of the colosseum, he is an archeologist, he studied the area of the Roman Forum while getting his degree. He was amazing!!

Highly recommend this tour company. It may be a bit more expensive, but well worth with the knowledgeable tour leaders!

I already paid to join the tour sponsored by Walks of Italy that Kathy gave a link to above. I printed the confirmation information I saw on the computer. Does this take away my need to buy a separate entrance ticket?

You would need to ask them that question. I don't know what's included with the tour.
