Cinque Terre to Rome experiences??

We're planning on flying into Florence for 5 days then moving to Cinque Terra (Vernazzo) for 2 nights then down to Rome for 4 days. Just doing initial research and planning. Has anyone made the trip from Cinque Terra to Rome by rail and if so how much time should i allot?

You can take a Frecciabianco high-speed train in 3-3.75 hours from La Spezia. It will take a few minutes to get to La Spezia from whatever CT town you are in. You change in La Spezia and your choices of departure are 08:20, 13:14, 16:20, 18:20 and one later, one even earlier. Very nice trip actually, with some Mediterranean views as you move down the coast.

As Laurel mentioned, there are a few direct trains from La Spezia Centrale to Roma Termini every day with a travel time of about 3H:45M. The direct trains will either be FrecciaBianca or InterCity both of which require train, date and time-specific reservations.

It's only a few minutes via local Regionale trains from Vernazza to La Spezia so it will be an easy connection. There are also numerous other trains on the La Spezia-Rome route each day which have one or more changes. I'd recommend using the direct trains if possible. You can see all the options on the Trenitalia website.

On the trip from Florence to Vernazza, you'll most likely have at least one or two changes depending on which train you choose.

We are planning to do the same thing. About how much would this cost per person? I have read to buy tickets in advance but how for in advance?

The base full price for a one way ticket from Spezia to Rome with an Intercity is €41, with a Frecciabianca it's €54. You can buy discounted tickets about 120 days prior. Savings range from 20% to 40% however a discounted ticket is not refundable and non changeable! therefore you must be reasonably sure of your travel plans. To the above add a few Euro for the portion from the Cinque Terre to Spezia. Note that if you stay in Monterosso some of those faster trains also stop there, and therefore if you board in Monterosso you don't need to change in Spezia.
