We are going to Italy in June. Staying in Florence 3.5 days. Thinking of doing a day trip to Cinque Terre. I have looked at the different options. From going with a tour 80 Euros p/p, to do it alone, which I am not really sure, don't want to rent cars or get lost!
Any suggestions? Also which will be the website to make reservations to the Vatican and Colosseum?

Getting lost is not usually an issue in the small villages of the Cinque Terre, especially if you navigate by the coastline. Most train departures between Firenze SMN station and Vernazza require connections at both Pisa and La Spezia, and take 3.5 hours each way, but the stations are small, making connections pretty smooth. How to Look Up Train Schedules Online gives you the DB train schedule link and tips for using it, with regional Italian train schedules published through June 14.
