We will be traveling to Cinque Terre (Vernazzo) the end of April from Florence. Can anyone tell me the best way to get there (car, train) and then our next destination is Venice. Is a car best? Should we go back to Florence and take a train to Venice?

You can get there by car or rail. The car is however not needed while at the Cinque Terre, therefore if you don't have a car already, don't bother to rent one to go there. From 5Terre to Venice you have two possible routes via rail. One is via Genoa/Milan, the other is via Pisa/Florence. Duration of travel varies depending on train connections etc. Use Trenitalia.com for schedules.
