We are 3 adults and plan to stay one night in Chianti region. Any recommendations will be helpful. We would like to take a tour from Florence or Sienna, stay back that night if possible and return the next day.

We found a small tour which visits vineyards and villages in chianti and was perfect for us - http://www.slow-days.com
They pick up from Florence.

Artviva.com does a full day tour, (Best of Tuscany Tour) which may be more than you are looking for, but it includes the winery tour, tasting, lunch and a visit to three hill towns. They also offer other, shorter tours focusing on the winery and tasting. They are based in Florence.
