We (2 adults) intend to rent a car from Venice to Rome with stops in between (Le Marche, etc.).
First: Best car rental company? We've had good luck with Sixt in Scotland but what about DiscoverCar? Others?
Second: Best location to pick up car in Venice (on the island or mainland?)
Third: Best location to drop off car in Rome (we're staying in the Trastevere area)

We always book our cars with AutoEurope.com- a US based consolidator with excellent customer service and usually best rates. They deal with all the major agencies- Hertz, Europcar, Avis, Sixt, etc
Have never heard of DiscoverCars
We always get No Deductible, full insurance so we don’t worry about filling up or any dings or scratches- Just drop the car and walk away. For us the few extra euros are worth it for peace of mind.
Make sure all drivers have an IDP-International Driving Permit- required by law and we have been asked for it. Get it at AAA before you leave US.
Do your homework regarding ZTLs, parking, speed cameras, tolls.
You can pick top car at Piazzale Roma and drop at Tiburtina.
Be very careful driving over the bridge when you leave P Roma- speeding tickets are very common there
Are you interested in renting a car for the day and using it to sightsee as you drive to Rome or are you spending nights on the way from Venice to Rome?
Driving straight through is probably a realistic 6-7 hours sticking to the freeways. If you add smaller side roads and sight seeing it will probably be a 10-12 hour trek. Compared to the 3.5-4 hours on the train for the same trek I don't know if I would consider it worth the hassle and expense. You could stop in Ferrara, Bologna or Florence (booking two separate tickets) for several hours of sightseeing and still beat the direct fastest driving time.
If you're planning on a few days of driving and seeing different towns on the way then I think it sounds much more interesting.
