We are planning on renting a car in Siena to explore the hill towns in the surrounding area and then the next day drive ourselves over to Lucca and drop the car off. (I am aware of the extra license we need to get from AAA)
Looking for suggestions on the towns we 'shouldn't miss' while having the car and we are also planning on making stops along the way to Lucca. As a note, we are doing a tour the day before getting the car up into Chianti for a couple wineries and San Gimignano.
Thanks!
You'll be driving right past San Gimignano but since you'll already have visited you won't stop. There are a few places worth visiting it just depends on your route. You can visit Pistoia, Montecatini Terme or Monteriggioni. All are worth a stop.

We're planning to rent a car in Siena as well, and drive to Montepulciano for a few nights. Any recommendations on car rental agencies in Siena? Any advice for driving the countryside? Have read a few horror stories about speed traps, traffic tickets that come months later and even car rental damage that can't be substantiated.
Don't know about the traffic infraction and damage allegations out there. We are purchasing extra coverage for the car through our travel insurance company. It's $9 more to our travel coverage.
Been looking at Europcar.com for the rental.
Planning on paying the extra 15€ for GPS.
We rented through the consolidator AutoEurope which is part of Kemwel. I had originally set this up for the Florence Airport. I called the Kemwel customer service and the guy was great about changing it to Siena and gave us the same price that included the super Collision damage coverage(walk away coverage).
Pick up location for our reservation is Siena City Office, 23/25 Viale Europa
Siena, Italy

I cannot use autoeurope.com, since we want to drop the car off in Lucca. It shows that isn't an option.
