We spent a week in Monopoli in 2012. The area is quite widespread so unless you spend all of your time at the hotel/resort, yes you will need a car. This region does not have a lot of tourist infrastructure. You are on your own to figure things out. We drove to Lecce, Ostuni, Aberobello and Matera on separate day trips. We wished that we had more time to explore to the north of us. At the marina in Monopoli you can buy fresh shrimp if you have a kitchen. The fishermen were very friendly. We really enjoyed these places and the people of Puglia. The food was very good too. Enjoy your trip.
You will use your time much more efficiently with a car. Matera is only an hour away from Monopoli. Otranto, Lecce, Ostuni, Martina Franca are some daytrips to consider.
