Is there a limit on how much you want to spend?
How far away are you staying from the bus/train stations?
Using taxis you can get to and from the bus/train stations with ease.
Maybe someone with UBER experience can advice you if that is an option.

STEP 1. Taxi from Rome Hotel to Rome Tiburtina bus station
STEP 2. Bus from Rome to Siena: http://en.baltour.it/Home/455-1-en.html
STEP 3. Taxi from Siena bus station to Siena hotel (unless hotel is short walking distance.

The train option is not as convenient as going to Siena requires changing from train to bus (or to another train) in Florence or Chiusi, which means having to lug your own bags off and on trains by yourselves. With the bus, the luggage is checked in and loaded in the belly of the bus by the driver, and you don’t have to worry about it until arrival in Siena.
