We will have 4 days in late August with a rental car to travel between Bologna and Florence. Does anyone have suggestions on must-see destinations, lodging, best/worst routes, etc?

I'm not exactly sure what you are asking but if you mean you have 4 days to get form Bologna to Florence that's about 3 day 22 hours too long.

We have four days to spend between Bologna and Florence. We are looking for suggestions on where to visit/spend the time - San Marino, Assisi, Lucca, Sienna etc.

If starting in Bologna, you could drive west to Parma and then take Route SS62 between Parma and Le Spezia. Route SS62 is extremely scenic, going through a national park. Then head back east to Florence, stopping in Lucca on the way. With a car, you can stay in agriturismos or country houses. Also recommend stopping at Villa San Donnino a country estate in Modena where they make excellent balsamic vinegar. Reservations needed but then an excellent free talk and tour are included. You could also stop at the Ferrari factory west if Modena. They have a good museum.
