What time of year and how many people? These factors can greatly affect the answers.
The best is subjective but certainly train for Florence > Arezzo > Venice (via Florence) is the most straightforward, easiest and probably the fastest. Certainly the fastest from Florence to Venice.
As you head into the Dolomites sometimes a car may be best - I'll leave that to someone else to answer.

You’ll need to be in Venice night before departure so I would change the order
Arezzo > Cortina > Venice
Eliminates that split stay
Probably pick up car as you leave Arezzo, drop it in Venice

curious as to where cardiff is, in wales or USA? how many people is we, what are your dates, do you have you accomodations booked? give us more info, budget in euros, what your plans are, any mobility issues, etc
i agree with ChristineH. venice should be your last stop and fly "home" from there. be very careful with your flight home if to USA. many flights leave at 6am. getting up at 3am, morning ritual and transportation to airport "VCE" (europeforvisitors.com)
if you are renting a car, anyone driving will need an IDP (aaa.com/IDP) which is mandatory in italy, get in USA before trip, plus read rules, laws, signage and especially ZTL's (no entrance areas unless you got deep pockets for a fine$$$!
people here on forum are here to help you with good bad & ugly without stress and great ideas for you to investigate. ask more questions and you decide what is best & easiest for "we". have fun and enjoy

Cortina has no train service - the closest stations are Calalzo to the south and Dobbiaco to the north, and the Dobbiaco line is partially closed this year for renovation. You either get there by bus - this is time consuming, and you have no time - or by car.

We will just be staying in Florence for the nite as our goal is to get to Areezo for 6 days. We do want a car in Tuscany.
If pick up car at Venice, do we get at airport or train station. The reason I ask is if we drive car back to Florence thinking will need to drop off near train station that we will take to Venice.
Perhaps we just drive to Venice.

When you reserve the car, you may have a choice of where you want to pick it up, between train station and airport. Airports are usually more expensive and outside the city, so it depends how convenient it will be for you to pick up and drop off at the locations. I would look at the car rental place you will use and see what options they offer. Sometimes different companies offer different locations, so you might want to compare a couple. I usually use Autoeurope to rent cars. Then I compare pick up and drop off locations and see how it fits best (for example, this summer I wanted to rent in Ravenna and drop in Como, but Avis does not offer this option, so instead, I am renting in Bologna and dropping in Como, which they did offer. In Bologna, I had the choice of picking up at the airport or train station).
You can pick car up in Florence at train station or once you get to Arezzo
I would get in Florence and just drive to Arezzo
Assuming you have oarking luned up in Arezzo?
Unfortunately you can’t use a car in Venice so either drop it there at P Roma then pick up another for Dolomites
Or pay to park it for 3 days
Drop it on return to Venice (airport if you are staying there night before departure or P Roma if you are staying in Venice proper)
