We (family of 4) will be traveling to Siena from Milan for the Palio in June/July. What is the best route to take on the train? Also, would renting a car? hiring a cab? be better?

Depends on what your plans are while in Tuscany. If you want to tour the countryside a bit and visit some vineyards, then a car would be the best option. Since there are four in your group it may even be cheaper than paying for four train tickets.
Use the link here on Rick Steves' website for the Duetsche Bahn and plug in Milano and Siena...the website will give you times and routes. Click on the names of the cities after the times come up and it will break it down into the stops the trains make. You could also go to the trenitalia website as well. I think it is www.trenitalia.it or www.trenitalia.com but I'm not absolutely certain. The link to Deutsche Bahn covers all the rails in Europe and the link from RS gets you to an english website.
