We will be in Rome through 4/1/12 and plan to either take a train or bus to towns in the Tuscany area. Which town would be the best to serve as a base to travel to surrounding areas. We would prefer to use public transportation rather than drive. Would enjoy biking in the areas as well.

Florence or Siena will likely give you the best options for access. As I sure you know, though, Tuscany is really best traveled by car. I've done is by train and bus (on my first trip) and it was great, until I went back and got a car. Then, no comparison. I'd consider picking up a car for a few days while there.
